چکیده
Strains of the food-borne pathogen Listeria (L.) monocytogenes have diverse virulence potential. This study focused on three outbreak strains: CC1 strain PF49 (serovar 4b) from a cheese-associated in Switzerland, clinical CC2 F80594 4b), and G6006 (CC3, serovar 1/2a), responsible for large gastroenteritis USA due to chocolate milk. We analysed genomes characterized vitro vivo. Whole-genome sequencing revealed high conservation major genes. Minor deviations gene contents were found autolysins Ami, Auto, IspC. Moreover, different ActA variants present. Strain showed prolonged survival liver infected mice. Invasion intracellular proliferation similar all strains, but strains increased spreading intestinal epithelial Caco2 cells compared G6006. Overall, this long-term 4b Future work will be needed determine genes molecular mechanism behind L. organs.

Genome Sequencing of Listeria monocytogenes “Quargel” Listeriosis Outbreak Strains Reveals Two Different Strains with Distinct In Vitro Virulence Potential
A large listeriosis outbreak occurred in Austria, Germany and the Czech Republic in 2009 and 2010. The outbreak was traced back to a traditional Austrian curd cheese called "Quargel" which was contaminated with two distinct serovar 1/2a Listeria monocytogenes strains (QOC1 and QOC2). In this study we sequenced and analysed the genomes of both outbreak strains in order to investigate the extent ...
متن کاملGenome comparison of Listeria monocytogenes serotype 4a strain HCC23 with selected lineage I and lineage II L. monocytogenes strains and other Listeria strains
More than 98% of reported human listeriosis cases are caused by specific serotypes within genetic lineages I and II. The genome sequence of Listeria monocytogenes lineage III strain HCC23 (serotype 4a) enables whole genomic comparisons across all three L. monocytogenes lineages. Protein cluster analysis indicated that strain HCC23 has the most unique protein pairs with nonpathogenic species Lis...
متن کاملMulti-virulence-locus sequence typing identifies single nucleotide polymorphisms which differentiate epidemic clones and outbreak strains of Listeria monocytogenes.
A recently developed multi-virulence-locus sequence typing (MVLST) method showed improved discriminatory power for subtyping genetically diverse Listeria monocytogenes isolates and identified epidemic clone II isolates associated with two recent U.S. multistate listeriosis outbreaks. To evaluate the ability of MVLST to distinguish other epidemic clones and outbreak strains of L. monocytogenes, ...
متن کاملDairy farm reservoir of Listeria monocytogenes sporadic and epidemic strains.
Identifying the reservoirs of a pathogen is vital for control of sporadic disease and epidemics. Listeria monocytogenes is a zoonotic foodborne pathogen that is responsible for 28% of food-related deaths in the United States annually, as well as a major cause of massive product recalls worldwide. To examine the role of the dairy farm as a potential source or reservoir for L. monocytogenes subty...
متن کاملRibotypes and virulence gene polymorphisms suggest three distinct Listeria monocytogenes lineages with differences in pathogenic potential.
A total of 133 Listeria monocytogenes isolates were characterized by ribotyping and allelic analysis of the virulence genes hly, actA, and inlA to uncover linkages between independent phylogenetic and specific virulence markers. PCR-restriction fragment length polymorphisms revealed 8 hly, 11 inl4, and 2 actA alleles. The combination of these virulence gene alleles and ribotype patterns separat...
